Oxidase test

Purpose
This test is useful in distinguishing bacterial genera.
Materials
- Filter paper
- 1% tetra methyl-p-phenylenediamine dihydrochloride solution.
- 24-hour culture
Instructions
- Cut filter paper into strips.
- Saturate filter paper strips with 1% tetra methyl-p-phenylenediamine dihydrochloride solution.
- Rub a platinum loopful or a sterile toothpick of a 24-hour culture onto the filter paper.
Expected results
A positive reaction is a rose-pink color that develops in 10 seconds.
Notes
- Be sure to use positive and negative controls.
- This test is very quick and easy to perform.
- Saturated filter paper strips can be stored in vials in the freezer until future use.
