Oxidase test

Purpose

This test is useful in distinguishing bacterial genera.

Materials

  • Filter paper
  • 1% tetra methyl-p-phenylenediamine dihydrochloride solution.
  • 24-hour culture

Instructions

  1. Cut filter paper into strips.
  2. Saturate filter paper strips with 1% tetra methyl-p-phenylenediamine dihydrochloride solution.
  3. Rub a platinum loopful or a sterile toothpick of a 24-hour culture onto the filter paper.

Expected results

A positive reaction is a rose-pink color that develops in 10 seconds.

Notes

  • Be sure to use positive and negative controls.
  • This test is very quick and easy to perform.
  • Saturated filter paper strips can be stored in vials in the freezer until future use.
