Yeast dextrose carbonate agar

Purpose

This medium is useful in identifying Xanthomonas species. Xanthomonas colonies are mucoid, convex, and shiny on YDC agar

Ingredients

Ingredient 1 L 500 mL
Distilled water 1 L 500 mL
Dextrose 20 g 10 g
Yeast extract 10 g 5 g
CaCO3, light powder 20 g 10 g
Agar 20 g 10 g

Instructions

  1. Autoclave dextrose separately in half of the distilled water
  2. Autoclave other ingredients in the other half of the distilled water
  3. Combine solutions. Stir before pouring plates so that CaCO3 is evenly distributed.

Notes

  • It is important to try to keep the calcium carbonate suspended in the medium
