Tetrazolium medium

Purpose
Used to test for pathogenicity of the vascular bacterial pathogen, Ralstonia solanacearum
Materials
| Ingredient | 500 mL | 1 L |

| distilled water | 500 mL | 1 L |
| peptone | 5 g | 10 g |
| casein hydrolysate | 0.5 g | 1 g |
| glucose | 2.5 g | 5 g |
| agar | 8.5 g | 17 g |
| triphenyl tetrazolium chloride | 0.025 g | 0.05 g |
Instructions
- Mix
- Autoclave
- Pour and cool
Expected results
- Two colony types appear on this medium. The nonpathogenic or weakly pathogenic colonies are round butyrous, deep red with narrow bluish borders. The highly pathogenic colonies are irregularly round, fluidal, white colonies or white colonies with pink centers. Clavibacter will not grow on this medium.
