Sucrose peptone agar

Purpose

This medium is a nonselective medium that is useful for general purpose bacterial isolations; however, it is not recommended for culturing Agrobacterium. It is particularly useful when looking for levan-producing pseudomonads. Xanthomonas campestris is characteristically mucoid on this medium. The clarity of this medium makes it useful for plate counts.

Ingredients

Ingredient 1 L 500 mL
distilled water 1 L 500 mL
sucrose 20 g 10 g
peptone 5 g 2.5 g
K2HPO4 0.5 g 0.25 g
MgSO4•7H2O 0.25 g 0.125 g
agar 15 g 7.5 g

Instructions

  1. Combine ingredients
  2. Adjust pH to 7.2-7.4 with 40% NaOH
  3. Mix and autoclave
