Purpose

| Nicotinic acid | 0.5 g | |
| L-asparagine (anhydrous) | 3 g | |
| K2HPO4 dibasic powder | 2 g | |
| MgSO4 • 7H2O | 0.2 g | |
| Sodium taurocholate (Difco) | 2.5 g | |

| Tergitol 7 (sodium heptadecyl sulfate) | 0.1 ml | |
| 2% nitrilotriacetic acid in 1.46% KOH (20 g + 14.6 g KOH/L) | 10 ml | |
| 0.5% bromothymol blue (Na salt)1 | 9 ml | |
| 0.5% neutral red | 2.5 ml | |
| 1N NaOH (do not use if a ppt. forms) | 5 ml | |
| 1% thallium nitrate | 1.75 ml | |
| 0.33% CoCl2 (0.83 g/250 ml H2O) | 50 ml | |
| Part 3 | ||
| 1N NaOH | 20 ml | |
| MOPS3 | 9 ml | |
| 10% CaCl2•2H2O | 2.5 ml | |
| agar | 4 g | |
| sodium polypectate | 18 g ml | |
Instructions
- Heat dH20 from part 1 to 80-90°C.
- Add the remaining ingredients from part 1 in order, be sure each is dissolved before adding next.
- Mix together the ingredients from part 2 in a separate container, add in the order listed.
- Add the mixture to the hot medium from part 1 and then add the ingredients from part 3 in order.
- Autoclave, pour thickly.
Notes
- The sulfone form is not water-soluble.
