Lactose broth

Purpose
Used for detection of the presence of coliform organisms, as a pre-enrichment.
Ingredients

| Bacto beef extract | 1.5 g | 0.75 g |
| Bacto peptone | 2.5 g | 1.25 g |
| Bacto lactose | 2.5 g | 1.25 g |
Instructions
- Dissolve ingredients and dispense medium into test tubes containing an inverted Durham tube.
- Autoclave (air bubbles are expelled from the Durham tube during autoclaving).
- Inoculate tubes and place in a water bath at 35 or 44.5 °C.
- Observe at 24 and 48 hours for an air bubble in the Durham tube.
Notes
- The coliform group is Enterobacteriaceae that ferment lactose with gas formation within 48 hours at 35 °C. Fecal coliforms are detected by lactose fermentation on M-FC Broth (see standard methods).
