Erwinia amylovora selective medium

Purpose
To differentiate Erwinia amylovora.
Ingredients
| Ingredient | 500 ml |
|---|---|
| Distilled water | 380 ml |
| Sucrose | 160 g |
| Nutrient agar | 12 g |
| Crystal Violet, 0.1% in ETOH | 0.8 ml |
| 0.1% Cycloheximide | 20 ml |
Instructions
- Autoclave, cool, pour plates and allow to dry for 2 hours prior to use or storage.
- After streaking, incubate at 28 °C for 60 hours. Examine the surface of each bacterial colony at 15 or 30X magnification with the aid of an obliquely oriented light source. Erwinia amylovora colonies develop characteristic craters apparent after 48-72 hr, but are most distinct after 60 hr. Erwinia herbicola and some other saprophytic isolates can grow on this medium, but do not develop craters.
