Purpose
For doing counts of Sclerotia of Macrophomina phaseolina isolated from soil.
Ingredients
- soil
- #10 screen
- pint jars
- 100% clorox
- RB Agar
Instructions
- Screen soil through #10 screen.
- Weigh 5 grams of soil into pint jars.
- In blender, mix sample with 250 ml of 100% Clorox. Blend for 10 sec. Let stand 5 min. Repeat for 3 total blends.
- Wash sample on screen 25um/500 mesh (45um/350 mesh) for 2 min.
- Rinse sclerotia into 250 ml flask with 25 ml of water.
- Add 90 (120) ml RB agar to each flask. Swirl and pour into 6-7 (10) plates.
- Incubate at 33oC for 7 days, count colonies.
