Crystal violet pectin

Purpose
A media to determine the pectolytic activity of bacteria.
Ingredients
Main Recipe

| NaNO3 | 1.0 g | 0.4 g |
| 0.075% Crystal Violet in 95% EtOH | 1 mL | 0.4 mL |
| 1N NaOH | 4.5 mL | 1.8 mL |
| 10% CaCl2*2H2O (aqueous) | 1 mL | 0.4 mL |
| Bacto Agar | 2.5 g | 1.0 g |
| Na Polypectate | 9.0 g | 3.6 g |
Instructions
- Combine NaNo3 with water
- Heat until near boiling
- Add Crystal Violet, NaOH, and CaCl2
- Pour mixture into a blender
- While on low speed, add Bacto Agar and Na Polypectate
- Blend for ~ 10-15s
- Pour into an autoclavable bottle with adequate headspace
- spray with anti-foam (if available)
- Autoclave
- Pour into petri dishes immediately after removing from autoclave
Notes
- Spot inoculate bacterial colonies on media. If colonies appear concave in the media, bacteria are pectolytic.
