Purpose
Selective for Clavibacter.
Ingredients
Solution A
| Ingredient | 1 L |
|---|---|
| Difco nutrient broth | 8 g |
| Yeast extract | 2 g |
| K2HPO4 | 2 g |
| KH2PO4 | 0.5 g |
| Agar | 20 g |
Solution B
| Ingredient | 1 L |
|---|---|
| 10% glucose | 50 ml |
| 1M MgSO4 • 7H20 | 1 ml |
Solution C
| Ingredient | 1 L |
|---|---|
| 1% polymyxin B sulfate | 2.5 ml |
| cycloheximide | 0.4 g in 10 ml H2O |
| LiCl | 10 g in 50 ml H2O |
| chlorothalonil 75%* | 0.16 g in 10 ml H2O |
| *Add these ingredients to sterile water (do not autoclave) | |
Instructions
- Autoclave Solutions A and B separately.
- After autoclaving, mix Solutions A and B together.
- Add Solution C to the mixture of Solution A and B.
Notes
- chlorothalonil 75% is a fungicide available from agricultural chemical supply companies, such as Target Chemical.
