Purpose

This medium is used to aid in the differentiation of various genera in the Enterobacteriaceae. Bacteria that can utilize citrate as the sole carbon source and inorganic ammonium salt as the sole source of nitrogen grow on this medium.

Ingredients

Ingredient 1 L 500 ml
Distilled water 1000 ml 500 ml
MgSO4 • 7H2O 0.2 g 0.1 g
NH4H2PO4 1 g 0.5 g
K2HPO4 1 g 0.5 g
Sodium citrate 2 g 1 g
NaCl 5 g 2.5 g
Bacto agar 15 g 7.5 g
Bacto bromothymol blue 0.8 g 0.4 g

Instructions

  1. Medium is usually prepared as agar slants, inoculated by lightly streaking the slant.
  2. Incubate up to 4 days at 37 °C.

  • Bacteria that can utilize citrate as the sole carbon source and inorganic ammonium salt as the sole source of nitrogen grow on this medium.

Expected results

  • A positive reaction is indicated by growth with an intense blue color on the slant (alkalinity). A negative reaction is no growth with no color change; the medium remains dark green.
