Purpose
Semi-selective medium for Xanthomonas
Ingredients

| Pourite | 1 drop | 0.5 drop (10μl) |
| K2HPO4 • 3H2O | 2.62 g | 1.31 |
| KH2PO4 | 0.5 | 0.25 |
| MgSO4 • 7H2O | 0.25 g | 0.125 g |
| Boric asid | 1.5 g | 0.75 g |
| Mannose | 10 g | 5 g |
| Yeast extract | 0.1 g | 0.05 g |
| Agar | 12 g | 6 g |
| Part 2 | ||
| Nicotinic acid* | 100 mg* | 50 mg* |
| Nalidixic acid (sodium salt)** | 30 mg** | 15 mg** |
| Cycloheximide† | 200 mg† | 100 mg† |
Instructions
- After dissolving the ingredients from part 1 in the distilled water, autoclave for 15 minutes.
- Cool in a water bath to 45-50 °C.
- Add ingredients from part 2
- dissolved in 20 ml sterile distilled water
- dissolved in 1 ml of 0.1 M NaOH
†dissolved in 1 ml absolute methanol
Notes
- Cycloheximide inhibits fungi. Pourite is an anti-bubble agent.
