Purpose

Semi-selective medium for Xanthomonas

Ingredients

Ingredient 1 L 500 ml
Part 1
Distilled water 980 ml 480 ml
Pourite 1 drop 0.5 drop (10μl)
K2HPO4 • 3H2O 2.62 g 1.31
KH2PO4 0.5 0.25
MgSO4 • 7H2O 0.25 g 0.125 g
Boric asid 1.5 g 0.75 g
Mannose 10 g 5 g
Yeast extract 0.1 g 0.05 g
Agar 12 g 6 g
Part 2
Nicotinic acid* 100 mg* 50 mg*
Nalidixic acid (sodium salt)** 30 mg** 15 mg**
Cycloheximide† 200 mg† 100 mg†


Instructions

  1. After dissolving the ingredients from part 1 in the distilled water, autoclave for 15 minutes.
  2. Cool in a water bath to 45-50 °C.
  3. Add ingredients from part 2
  • dissolved in 20 ml sterile distilled water
  • dissolved in 1 ml of 0.1 M NaOH

†dissolved in 1 ml absolute methanol

Notes

  • Cycloheximide inhibits fungi. Pourite is an anti-bubble agent.
