Purpose

For isolating sclerotia of Macrophomina phaseolina from soil.

Ingredients

• Soil
• #10 screen
• 10% Clorox
• CMR agar

Instructions

1. Screen soil through #10 screen.
2. Weigh 5 grams of soil.
3. In blender, mix sample with 250 ml of 10% Clorox. Blend for 10 sec. Let stand 5 min. Repeat for 3 total blends.
4. Wash sample on screen (25um/500 mesh). Rinse sclerotia into 250 ml flask with a minimum of water.
5. Add 100 ml CMR agar to each flask. Swirl and pour into 6-7 (10) plates.
6. Incubate at 33°C for 7 days, count colonies.
