|---|---|---|
| Part 1 | ||
| Distilled water | 1000 ml | 500 ml |
| Hempseed | 50.0 g | 25.0 g |
| Part 2 | ||
| Agar | 17.0 g | 8.5 g |
Instructions
- Mix the ingredients in part 1 and let sit overnight
- After soaking broth overnight, autoclave for 15 minutes, then strain broth through cheesecloth. Add ingredient in part 2.
- Bring up to volume with distilled water. Autoclave for 15 minutes.
- To induce sexual reproduction of Phytophthora capsici and P. parasitica, inoculate with small disks of pairs of isolates about 15 mm apart and grow at 21°C for 5-7 days.
Notes
- Hempseed provides sterols necessary for sexual reproduction of oomycetes: however, vegetable-based media such as corn, oatmeal, lima bean, carrot, potato V8, etc., also provide sterols.
- Hempseed medium has the advantage of being very clear thus allowing for unobstructed viewing of structures through the bottom of a Petri dish.
