Esterase activity (Tween 80 Hydrolysis)

Purpose
Used to detect the ability of bacteria to hydrolyze the lipid, Tween 80.
Ingredients

| Distilled water | 1000 ml | 500 ml |
| Peptone | 10 g | 5 g |
| NaCl | 5 g | 2.5 g |
| CaCl2 • 2H20 | 0.1 g | 0.05 g |
| Agar | 15 g | 7.5 g |
Instructions
- Adjust pH to 7.4.
- Autoclave and add Tween 80 (autoclaved) at 10 ml/liter rate.
- Pour into Petri plates.
- Spot or streak bacterial isolates. Esterase will hydrolyze the tween and subsequent precipitation with the calcium chloride will appear as a turbid zone around the colonies.
