Esterase activity (Tween 80 Hydrolysis)

Purpose

Used to detect the ability of bacteria to hydrolyze the lipid, Tween 80.

Ingredients

Ingredient 1 L 500 ml
Distilled water 1000 ml 500 ml
Peptone 10 g 5 g
NaCl 5 g 2.5 g
CaCl2 • 2H20 0.1 g 0.05 g
Agar 15 g 7.5 g

Instructions

  1. Adjust pH to 7.4.
  2. Autoclave and add Tween 80 (autoclaved) at 10 ml/liter rate.
  3. Pour into Petri plates.
  4. Spot or streak bacterial isolates. Esterase will hydrolyze the tween and subsequent precipitation with the calcium chloride will appear as a turbid zone around the colonies.
