Purpose

For doing counts of Sclerotia of Macrophomina phaseolina isolated from plant material.

Ingredients

• Plant sample
• RB agar

Instructions

1. All flasks and media must be kept warm until plates are poured.
2. Check on correct sample weight for sample set.
3. Weigh out sample into 250 ml flask.
4. Add 100 ml RB agar to each flask.
5. Incubate at 45°C for 20 minutes.
6. Swirl and pour into approximately 6-7 petri plates.
7. Allow media to harden before moving.
8. Incubate in dark at 33°C for 7 days, count colonies
