Purpose

Ingredients

• 100 g soil
• 20 ml extract
• 0.1 M Tricine buffer pH 11.2

Urea	20	40	60	80
DMSO	20	40	60	80
Adenosine	4	8	12	16
EDTA	2	4	6	8

Instructions

1. Blend soil and extract for 20 seconds, 3 times.
2. Centrifuge at 3000 G in Sorval SS-1 at room temp. for 20 minutes.
3. Dilute 1:10 with Tricine buffer.
4. Check if pH is between 7 and 8
5. Place samples in freezer
6. Lubrol dissolve 0.5 g in 34 ml sterile H2O by warming hold at 35°C

Notes

• Add EDTA just before use
